Abstract

The organism that can live outside of the ‘normal’ range of at least one environmental factor is an extremophile. A thermophile is an organism a type of extremophile that thrives at relatively high temperatures, between 45 and 122 °C. There are several mechanisms used thermophiles hyperthermophiles to maintain the stability of the proteins for their survival. To study this stability there are many Bioinformatics tools available. The amino acid composition is believed to alter the intrinsic stability of proteins. Several comparative studies of the primary sequence obtained from 97 thermophilic organisms have been carried out between Mesophilic and Thermophilic Alpha-amylases to understand the amino acid composition for the thermostability of proteins. This work presents some generalized features of amino acid composition found in thermophilic proteins, including an increase in residue hydrophobicity, a decrease in uncharged polar residues, an increase in charged residues, an increase in aromatic residues. The differences of amino acids composition between thermophilic and mesophilic proteins are related to some properties of amino acids. These features provide guidelines for engineering mesophilic protein to thermophilic protein.
