Neuromodulatory Potency of Essential Oil in Cadmium Induced Cognitive Impairment on the Prefrontal Cortex of Wistar Rats

Abstract

Background: Cadmium (Cd) is an environmental trace element that severely affects the functionality of nervous system particularly the prefrontal cortex (PC).

Objective: The research accessed the role of clove essential oil (CLO) in Cd induced cognitive impairment on rats.

Methods: Twenty-five Wistar rats were randomly assigned into five groups of five rats each. Group A (Control) received water, group B received 20mg/kg of Cd only, C was given 10mg/kg of CLO, D was given 20mg/kg of CLO while E was given 20mg/kg of DMSO. All groups except A received 20mg/kg of Cd before treatment. Cognition was accessed using Novel Object Recognition Test (NORT) and Sociability Chamber Tests (SCT) while blood sample was used to estimate GST, GPx, GSH, and GOT enzymes.

Results: The result showed that GSH and GST was significantly decreased in group B compared to control and increased significantly in treated groups compared to group B (P≤0.001). Group B showed a reduced discriminative index (0.34±0.01) compared to A (0.60±0.04). While the reduction continued in groups C and E (0.14±0.09 and 0.27±0.02); group D increased (0.81±0.02). The SCT showed a significant increase in Time spent in common chamber (TSICC) in group B (23.50±6.51) compared to A (19.25±2.78) which was significantly increased in groups C and D while decreasing in E compared to B. Microscopically, group A showed normal histoarchitecture with several alterations on group B while treated groups showed significant improvement in cell regeneration.

Conclusion: Clove oil ameliorates cadmium toxicity by increasing antioxidant activity and increases cognition in rats.Background: Cadmium (Cd) is an environmental trace element that severely affects the functionality of nervous system particularly the prefrontal cortex (PC).
