Comparative Study of Two Acanthogobius Species Based on Genome-Wide Survey Analyses

Abstract
Abstract Acanthogobius stigmothonus and A. flavimanus are native to Asia and play an important role in the coastal ecosystem and economic development. However, the genomic data of them is lacking. In this study, we conducted genome survey analysis of two species using next-generation sequencing technology. The genome sizes were estimated to be 998.27 Mb, 978.96 Mb by using K-mer analyses for A. stigmothonus and A. flavimanus , respectively. The heterozygosity, repeat ratios, and numbers of microsatellite motifs all suggested that A. stigmothonus is closely related to A. flavimanu s at the genetic level. Moreover, the complete mitochondrial genome sequences were assembled from the whole genome data and the phylogenetic analyses genetically supported the validation of A. stigmothonus and A. flavimanus . The pairwise sequentially Markovian coalescent (PSMC) analysis revealed the trajectory of the demographic history across time. This study provides a large number of gene resources for further excavation and utilization of A. stigmothonus and A. flavimanus .
