Abstract
The use of small-molecule organic probes with near-infrared fluorescence in biology has gained momentum in recent years, and their numerous advantages have triggered new research and clinical applications. Among them, hemi-cyanine dyes are characterized by their structural diversity and excellent quantum yield, making them versatile for the development of optical probes. In this work, a novel hemi-cyanine dye containing a tetrahydropyridine ring was synthesized and a lysosomal target was introduced into its structure to create a pH-sensitive near-infrared fluorescent probe capable of successfully targeting lysosomes, enabling non-invasive detection of changes in the acidic environment of lysosomes in living organisms. The photophysical properties of the dye indicate that it has good NIR fluorescence properties and imaging studies have demonstrated its good imaging capability as an NIR fluorescent agent for imaging living cells. It is noteworthy that the N of the tetrahydropyridine ring of the hemi-cyanine dye with the new structure can serve as a new target for.
