Quantification of Vitamin B9(Folic Acid) & Vitamin B12(Cyanocobalamin) in Fortified Rice Kernel Using Lc-Ms/Ms: A Validation Study

Abstract
By creating a sample preparation technique and fine-tuning the analytical instrument parameters, the vitamin B9 and B12 content of Fortified Rice Kernel (FRK) was ascertained. Following the purification of the water-soluble extract using solid-phase extraction and a liquid chromatography tandem mass spectrometer (LC-MS/MS), vitamin B9 and vitamin B12 were extracted from spiked rice kernel. The sample extract was cleaned by centrifuging, filtering, gently passing through a cartridge, and diluting it with buffer. After solid phase extraction, the sample solution was injected into an LC-MS/MS device for measurement of folic acid and cyanocobalamin from a spiked rice kernel sample. This was done by serving Certified Reference Materials (CRM) and optimising the mobile phase for vitamin B9 and B12 detection.
