Abstract

Backgrounds: The prognosis of nasopharyngeal carcinoma (NPC) is often complicated by a highincidence of distant metastasis and local treatment failure, resulting in patient death. Our goal is to explore early prognostic biomarkers that can offer a time window for administeringpersonalized therapy tailored in advance to NPC patient at high risk of recurrence.

Methods: In a study involving 46 cases, laminin expression in cancer tissue and serumlaminin levels were investigated through immunohistochemistry and enzyme-linkedimmunosorbent assay, respectively. The patients were followed up for 5 years.

Results: Of particular significance were the findings pertaining to the recurrent and death group,where the rates of laminin staining (42.86%, 38.46%) were notably lower compared to theircounterparts (77.78%, 80%), respectively. Intriguingly, serum laminin concentrations in the recurrentand death group (0.688±0.225, 0.695±0.253) were higher than those in their counterparts. Thisdiscrepancy underscores the potential prognostic value of laminin expression in NPC.Notably, the concentration before radiotherapy (0.642±0.247g/L) surpassed that afterradiotherapy (0.235±0.221g/L), suggesting a potential therapeutic impact on laminin levels.Intriguingly, in the recurrent group, serum laminin elevation occurred approximately 2 monthspreceding the relapse confirmed by CT scan. This temporal association suggests the feasibility of utilizingserum laminin levels as an early indicator of disease recurrence.

Conclusion:the study elucidates the potential utility of laminin expression/serum levels as a valuablebiomarker for predicting illness progression and recurrence. Within a 2-month time window, high-riskNPC patients can receive personalized preventive therapy before the tumor becomes detectable on a CTscan.
