The G Protein-Coupled Estrogen Receptor and Tumor Microenvironment Score May Serve as a Prognostic Marker in Cervical Cancer
Objective: Cervical cancer (CC) is one of the most common gynecological cancers which needs reliable biomarkers to monitor the prognosis of CC. G protein-coupled receptor (GPR) has been reported to regulate the cell proliferation of various cancers. The tumor microenvironment (TME) is the hot spot for cancer therapy. However, the combination of GRP and TME to the prognosis of the CC has not been fully studied.
Methods: Transcriptome data for cervical cancer was collected from the TCGA database. The cohort was categorized into four groups with GFR and TME scores and compared to prognosis. Single-cell mRNA sequence (scRNA-seq) data from four CESC tumor samples were analyzed by the GPR-related score for cell-cell communication. CESC patients were further stratified into four groups based on GPR, TME, and tumor mutational burden (TMB), and overall survival was compared among these groups.
Results: With the high expression of GPR, the level of TME did not affect the results, however the low expression of GPR showed patients with high TME expression have a good prognosis and those with low TME expression have a poor prognosis. Then a total of 11 clusters were annotated and the pre-dendritic cells (pDC) had the highest GPR-score. Cell communication analysis showed that GFR-low pDCs and GFR-high pDCs were strongly associated with immune cells. And TMB_high group of patients seems had a better prognosis.
Conclusions: With the use of GFR and TME score we constructed, cervical cancer can be better to predict the prognosis and the possible immunotherapy method.
